Masry are second in the group with seven points, tied with Kaizer Chiefs, who sit in third.
Zamalek lead the group with eight points, while Zambia’s Zesco United are bottom with no points.
"The result of this match will determine which teams qualify for the quarter-finals, as our group is very competitive," Masry’s Tunisian coach Nabil Kouki said at the pre-match press conference.
Masry secured a 2-1 win over Kaizer Chiefs in the previous meeting, followed by a dramatic 3-2 victory away to Zesco United.
They also earned a goalless draw against Zamalek and suffered a 2-1 loss to fellow Egyptians, Zamalek.
With these results, they will be confident heading into Sunday’s fixture.
"We are aiming for a positive result, and the game will be more tactical. We know the strengths and weaknesses of Kaizer Chiefs’ squad," he added.
Masry will be without key defender Baher El Mohamady, who picked up an injury during the Zamalek match, while midfielder Bonheur Mugisha is suspended.
"I am saddened by El Mohamady’s absence through injury, but I am confident in the other players in the squad," Kouki said.
The Port Said-based club will need a victory to keep their hopes of advancing to the knockout stage alive.
