The Whites secured a 1-1 away draw in the first leg last week, leaving the tie finely poised ahead of the return in Cairo.
Zamalek will welcome back veteran defender Mahmoud Hamdi El-Wensh, who returns from a muscle injury that kept him out of the first leg.
Midfielder Seif Gaafar, goalkeeper Mohamed Awad and winger Ahmed Hamdi remain sidelined due to technical reasons.
Coach Motamed Gamal said on Saturday his side face a difficult task against a “strong” Otoho d’Oyo team despite the first-leg result.
“Things are in our favour on paper, but we shouldn’t depend on the first-leg result, especially as we will be facing a strong opponent,” the 53-year-old told a pre-match press conference.
“We will start the game strongly to make our mission easier,” he added.
Gamal said his team are well prepared for the match and praised the expected home support.
“We prepared in the best way and hope to be at our best to make our fans proud,” he said. “We played a friendly that helped us assess players returning from injury. Zamalek fans mean a lot to me and I feel calm with their presence in the stands.”
Otoho d’Oyo coach Sekou Seck said his side would fight to stay in the competition.
“We will play only to win. This is the only competition for us and we will do our best to remain in contention,” he said.
“The first-leg result wasn’t ideal and Zamalek were the favourites.”
The winners will face Algeria’s CR Belouizdad, who beat Masry, in the semi-finals.
