The attack occurred near the headquarters of Israel's Mossad spy agency, according to the Associated Press.
At least 37 people were wounded, including six in serious condition, and were initially transported to nearby hospitals, Israel's Magen David Adom emergency service said in a statement.
Police confirmed multiple injuries.
Officers and ambulances rushed to the scene, where Israeli television showed police cordoning off the area as medics tended to the injured. A helicopter hovered overhead.
Minutes after the attack, Israeli police announced that the truck driver who rammed into people outside Glilot Base in central Israel was shot and "neutralized" by armed civilians nearby.
According to an initial police investigation, a bus had stopped at the station outside the base to drop off passengers when the truck rammed into the stop and those waiting there.
Police said further details, including the motive, were under investigation.
