Ismaily's assistant coach Ashraf Khedr resigned from his post after the club's defeat on Saturday to Masry in the 13th week of the Egyptian Premier League.
"My decision is final and I thank all the team and board members," Khedr told the club's official website on Monday.
Khedr was assisting former coach Ahmed 'Mido' Hossam who left the team to lead his boyhood club, title-holders Zamalek.
Khedr worked as a caretaker coach for the coastal club until they signed Tunisian tactician Nasreddine Nabi.
"I led the team during the most difficult time in its history and I am happy with what I did because it was the best I could do in the tough circumstances," he added.
Former coach Mido, 32, began leading the Dervishes at the start of the season hoping to change the team's fortunes but a dispute with captain Hosni Abd Rabou led to his resignation.
"I hope the club support the new manager because he is capable of leading the team back to winning days," Khedr concluded.
After Sunday's game, Ismaily moved to eighth position in the table with 17 points from 11 matches. Port Said- based club Masry lead the table with 25 points from 12 matches.
The three-time Egyptian Premier League champions Ismaily failed to finish in the top three last season, meaning that they were ineligible to play in any African-based competition.
Nabi previously coached the AC Leopards of Congo before leading Saudi Arabian club Al-Hilal.
