Hamdi, who scored the winner in Zamalek's 1-0 victory over RS Berkane of Morocco in the second leg of the Confederation Cup final on Sunday, failed to complete the game and was substituted five minutes before the end of the first half.
The team's doctor initially described Hamdi's injury as serious, and on Tuesday, it was confirmed that he had indeed suffered an ACL tear.
The 26-year-old will undergo surgery abroad in the coming days.
Hamdi, a former Ahly midfielder, joined Zamalek from CF Montreal in January and made a significant impact, scoring twice in 12 appearances in all fronts with the White Knights.
He becomes the third Zamalek player this season to sustain an ACL injury, joining defender Mahmoud Hamdi and veteran fullback Mohamed Abdel-Shafi.
Zamalek's next fixture is a league game against Modern Future on Thursday.
