The win ended a 24-match winless streak against Zamalek, who had previously won 13 and drawn 11 of their encounters with Degla.
Zamalek took an early lead through Nasser Maher, who struck from inside the box just 10 minutes into the match. However, Wadi Degla responded strongly in the second half.
Ahmed Farouk levelled in the 60th minute and Mahmoud Diasti completed the comeback, 67, to seal the historical win.
Strong display
Zamalek went ahead in the opening 10 minutes with Maher capitalizing on a poor defending to score from close range.
The hosts reponded and controled the tempo in midfield but were wasteful in front of goal. Maher could have doubled his goal tally when he rounded his marker but his shot was blocked by a last minute tackle.
Degla maintained their attacking pressure in the second half and were ultimately rewarded with a historic result over the league contenders.
At the hour mark, Farouk leveled the score after a beautiful combination to tap home from a tight angle.
Diasti completed the comeback seven minutes later, converting from close range after a deflected ball fell into his path.
Aly Hussein could have make it 3-1 after a brilliant solo move that saw him go clear on target but his chip ball went over the bar.
Both teams ended the game with only 10 men on the pitch as defender Mahmoud ‘El-Wensh’ Hamdi was shown a straight red card in the 73rd minute and Ibrahim El Bahnasy was also dismissed for a second yellow card five minutes from time.
The result leaves Zamalek second in the standings with 10 points, one behind leaders Masry. Wadi Degla climb to 10th place with seven points.
Zamalek, who finished third last season, return to action on 14 September against Masry in the sixth matchday following the international break.
