Tunisian striker Seifeddine Jaziri put Zamalek ahead with a goal just four minutes into the match.
However, Kaizer Chiefs equalized in injury time, thanks to a mistake from Zamalek goalkeeper Mohamed Sobhi.
Zamalek, who kicked off their group stage campaign with a win over Zambia's ZESCO United last Sunday, are second in Group D with four points, two behind leaders Masry.
Chiefs are third with one point, while ZESCO remain at the bottom with no points.
The visitors started the match strongly, applying early pressure despite playing away.
Jaziri, 32, capitalized on a well-delivered free-kick cross, firing a powerful shot from inside the box to give Zamalek the lead in the fourth minute.
Kaizer Chiefs came close to equalizing just three minutes later, but Gastón Sirino’s powerful strike from outside the area was denied by the left post.
Jaziri thought he had scored again in the 10th minute, but his effort was ruled offside.
The hosts dominated possession for much of the first half, although Zamalek's defense held firm to keep them at bay.
Kaizer Chiefs continued to apply pressure after the break but were unable to break down Zamalek's backline, with the visitors rarely threatening on the counter.
The fourth official added eight minutes of injury time, and in the dying moments, Kaizer Chiefs capitalized on a Zamalek defensive error.
Dillan Solomons sent a wide cross from the right wing into the box, and Sobhi could only deflect it into his own net, securing the draw for the hosts.
