Gerard Pique will also be out as the centre back continues his recovery from a leg injury.
Barcelona coach Pep Guardiola said on Monday that “the doctors have told me that if (Puyol) is fine today he will be back tomorrow.”
The 33-year-old Puyol has been struggling with lower body injuries and has made limited appearances for the defending champions since last winter.
Barcelona are third place in the Spanish league after drawing 0-0 with Sevilla.
